A general framework by which dynamic interactions within a protein will promote the necessary series of structural changes, or “conformational cycle,” required for function is proposed. It is suggested that the free‐energy landscape of a protein is biased toward this conformational cycle. Fluctuations into higher energy, although thermally accessible, conformations drive the conformational cycle forward. The amino acid interaction network is defined as those intraprotein interactions that contribute most to the free‐energy landscape. Some network connections are consistent in every structural (...) state, while others periodically change their interaction strength according to the conformational cycle. It is reviewed here that structural transitions change these periodic network connections, which then predisposes the protein toward the next set of network changes, and hence the next structural change. These concepts are illustrated by recent work on tryptophan synthase. Disruption of these dynamic connections may lead to aberrant protein function and disease states. (shrink)

A novel Protocol Ethics Tool Kit (‘Ethics Tool Kit’) has been developed by a multi-stakeholder group of the Multi-Regional Clinical Trials Center of Brigham and Women9s Hospital and Harvard. The purpose of the Ethics Tool Kit is to facilitate effective recognition, consideration and deliberation of critical ethical issues in clinical trial protocols. The Ethics Tool Kit may be used by investigators and sponsors to develop a dedicated Ethics Section within a protocol to improve the consistency and transparency between clinical trial (...) protocols and research ethics committee reviews. It may also streamline ethics review and may facilitate and expedite the review process by anticipating the concerns of ethics committee reviewers. Specific attention was given to issues arising in multinational settings. With the use of this Tool Kit, researchers have the opportunity to address critical research ethics issues proactively, potentially speeding the time and easing the process to final protocol approval. The digital shift leads to increasing changes. Employees can deal with changes through informal learning that enables needs-based development. For successful informal learning, self-regulated learning is crucial, i.e., to set goals, plan, apply strategies, monitor, and regulate learning for example by applying resource strategies. However, existing SRL models all refer to formal learning settings. Because informal learning differs from formal learning, this study investigates whether SRL models can be transferred from formal learning environments into informal work settings. More precisely, are (...) all facets relevant, and what are the relational patterns? Because informal workplace learning occurs through interaction with the context, this study investigates the influence of context, i.e., organizational learning culture and job characteristics on SRL. Structural equation modeling of N = 170 employees in various industries showed the relevance of the self-reported metacognitive strategies planning, monitoring, and regulation; the resource strategies help-seeking and effort regulation; and deep processing strategy elaboration. However, there was no evidence for organization strategies. The learning strategies were associated with self-efficacy and mastery-approach goal orientation. Regarding context, results supported indirect effects over self-motivational beliefs of learning strategies. Organizational learning culture was connected with mastery-approach goal orientation, whereas job characteristics autonomy and feedback were related to self-efficacy, which were again related to SRL strategies. Therefore, context can empower employees not only to accomplish their tasks but to develop themselves by applying SRL strategies. The results are discussed, and practical implications are outlined. BackgroundStory recall tests have shown variable sensitivity to rate of cognitive decline in individuals with Alzheimer’s disease biomarkers. Although SR tasks are typically scored by obtaining a sum of items recalled, item-level analyses may provide additional sensitivity to change and AD processes. Here, we examined the difficulty and discrimination indices of each item from the Logical Memory SR task, and determined if these metrics differed by recall conditions, story version, lexical categories, serial position, and amyloid status.Methodsn = 1,141 participants from (...) the Wisconsin Registry for Alzheimer’s Prevention longitudinal study who had item-level data were included in these analyses, as well as a subset of n = 338 who also had amyloid positron emission tomography imaging. LM data were categorized into four lexical categories, and by serial position. We calculated difficulty and discriminability/memorability by item, category, and serial position and ran separate repeated measures ANOVAs for each recall condition, lexical category, and serial position. For the subset with amyloid imaging, we used a two-sample t-test to examine whether amyloid positive and amyloid negative groups differed in difficulty or discrimination for the same summary metrics.ResultsIn the larger sample, items were more difficult in the delayed recall condition across both story A and story B. Item discrimination was higher at delayed than immediate recall, and proper names had better discrimination than any of the other lexical categories or serial position groups. In the subsample with amyloid PET imaging, proper names were more difficult for Aβ+ than Aβ−; items in the verb and “other” lexical categories and all serial positions from delayed recall were more discriminate for the Aβ+ group compared to the Aβ− group.ConclusionThis study provides empirical evidence that both LM stories are effective at discriminating ability levels and amyloid status, and that individual items vary in difficulty and discrimination by amyloid status, while total scores do not. These results can be informative for the future development of sensitive tasks or composite scores for early detection of cognitive decline. (shrink)

We examine the sequences A that are low for dimension, i.e. those for which the effective dimension relative to A is the same as the unrelativized effective dimension. Lowness for dimension is a weakening of lowness for randomness, a central notion in effective randomness. By considering analogues of characterizations of lowness for randomness, we show that lowness for dimension can be characterized in several ways. It is equivalent to lowishness for randomness, namely, that every Martin-Löf random sequence has effective dimension (...) 1 relative to A, and lowishness for K, namely, that the limit of KA/K is 1. We show that there is a perfect [Formula: see text]-class of low for dimension sequences. Since there are only countably many low for random sequences, many more sequences are low for dimension. My review of Cornelius Castoriadis' book Crossroads in the Labyrinth ended with the apt reference, I now see, to the emperor being naked. In Joel Whitebook's second review, largely irrelevant to my criticisms of Castoriadis, he fears, though he doesn't know me personally, that only the lack of psychological counseling can explain my uncontrolled anger against Castoriadis. Let me dignify his long distance psychoanalysis by passing over it in silence. Silence is also the best remedy for Whitebook's transcendental deduction that (...) I have a “pluralistic, localistic, discontinuous ontology.” Even if I knew what that was I find it amazing that he located it in a 10 page review. Perhaps Whitebook just “feels” the presence of this ontology along with evidence of my mental problems. (shrink)

, Lawrie Reznek argues that disease is not a natural kind term. I raise objections to Reznek's two central arguments for establishing that disease is not a natural kind. In criticizing his a priori, conceptual argument against naturalism, I argue that his conclusion rests on a weaker argument that appeals to the empirical diversity in the symptoms and manifestations of disease. I also raise questions about the account of natural kinds which Reznek utilizes and his point that conventions for classification (...) are excluded by there being natural kinds. From 1969 through the 70's Mitchell Franklin was Emeritus Professor of Law and Philosophy at SUNY Buffalo. Over this period his teaching gradually shifted to philosophy where he gave a series of lectures on Hegel, Marx and Neo-Hegelianism, which attracted and influenced a new group of students. These philosophy students were rediscovering the Continental tradition and turning to phenomenology, Western Marxism and German Idealism against die positivist and analytic traditions which had a dying but tenacious hold on philosophy. The article analizes the several times of Proclus‘s reception by Nicholas of Cusa’s thought. The direct reading of Proclus can be established because Expositio in Parmenidem Platonis –Cod.Cus. 186– and Elementatio theological –Cod.Cus.195– (Moerbeke’s translation) and De theologia Platonis Libri VI –Cod.Cus.185– (Petrus Balbus’s translation) are in his Library in Bernkastel-Kues with his marginalia. The assimilation of doctrines can be considered assuming that the implicits and explicits references to Plato’s Diadochus, especially in the last works.

Contemporary Continental Philosophy steps back from current debates comparing Continental and analytic philosophy and carefully, yet critically outlines the tradition’s main philosophical views on epistemology and ontology. Forgoing obscure paraphrases, D’Amico provides a detailed, clear account and assessment of the tradition from its founding by Husserl and Heidegger to its challenge by Derrida and Foucault. Though intended as a survey of this tradition throughout the twentieth century, this study’s focus is on the philosophical problems which gave it birth and even (...) now continue to shape it.The book reexamines Husserl as an early critic of epistemological naturalism whose grasp of the philosophical importance of the theory of meaning was largely ignored. Heidegger’s contrasting effort to revive ontology is examined in terms of his distinction between ontic and ontological questions. In contrast with many earlier studies, the author outlines confusions engendered by the misappropriation of the distinct philosophical agendas of Husserl and Heidegger by such famous figures as Sartre and Merleau-Ponty. The book is also original in its emphasis on how social externalism in epistemology, inspired by Karl Mannheim, influenced this tradition’s structuralist and Marxist phases. The philosophical defenses of a theory of interpretation by Gadamer and Habermas are closely examined and assessed and the study concludes with a a probing yet balanced account of Foucault and Derrida as critics of philosophical autonomy. The book concludes by reassessing this century-long divide between the analytic and Continental traditions and its implication for the future of philosophy. (shrink)

This article studies the underinvestigated but fascinating issue of the sociological determinants of accounting misbehavior while focusing on an allegedly illicit accounting practice (i.e., restatement) in family- vs. nonfamily-controlled corporations. Under the framework of institutional anomie theory, we examined whether sociological structures (i.e., legal forces and cultural values) influence accounting errors inducing restatements. By applying a multivariate regression analysis to a sample of restating firms listed in 23 countries during the 2006 to 2014 period, we found that legal forces and (...) cultural values significantly moderate the severity of accounting errors. The results of this study suggest that investors, managers, and policymakers should more fully consider the sociological structures of societies when debating the feasibility of corporate misbehaviors, as combining firm-level and country-level analyses could help to predict a firm’s accounting misbehaviors, such as more severe accounting errors. ExcerptDavid Graeber and David Wengrow, The Dawn of Everything: A New History of Humanity. New York: Farrar, Straus and Giroux, 2021. Pp. 704. The Dawn of Everything is not just a massive book in terms of its total number of pages but also in the amount of archaeological evidence discussed concerning human “prehistory.” The authors range over current disputes within their disciplines as well as discussing in some detail political philosophies of the seventeenth and eighteenth centuries. In spite of its (...) size and range, the book has been widely reviewed and has had a publication success rarely seen with academic works. Attention to it may be in part due to the publishing career of its co-author David Graeber, who tragically died in 2020 just before the book appeared in print. The reason I mention this point is that Graeber’s work had both an academic and non-academic audience, in such books as Bullshit Jobs: A Theory, Debt: The First 5,000 Years, and The Democracy Project: A History, a Crisis, a Movement. He also had a public profile during the Occupy Wall Street movement beginning in 2011, which highlighted his advocacy of anarchism. Graeber’s political philosophy plays a significant role in this book. (shrink)

Un ejemplo recurrente es el cogito cartesiano: ¿por qué es verdaderamente una creación? En primer lugar porque no supone otros conceptos pero, sin embargo, es comprensible porque está sostenido por una (...) imagen presupuesta: todo el mundo sabe qué significa pensar. Con todo, sostienen los autores, "hay algo a medio camino entre el plano del concepto y el plano preconceptual, que va de uno a otro" (DELEUZE; GUATTARI, 1993, p 63-ss). Es el personaje conceptual. En el caso del cogito, sostienen los autores, se trata del Idiota: él es quien dice Yo, él es quien lanza el cogito... Esta revisión tiene lugar a la luz del De pace fidei de Nicolás de Cusa, que ha sido interpretado en la modernidad como un modelo de tolerancia religiosa. La inaccesibilidad de la verdad o deus absconditus y el factum de la diversidad, sientan las bases para la búsqueda de la unidad en la diferencia. La fundamentación filosófica de la Trinidad y la Encamación, cuya aceptación (...) es indispensable para la construcción de la paz de la fe, muestra la coherencia que Nicolás establece entre filosofia y teologia, entendidas ambas en el sentido de la docta ignorantia. At the end of World War II Karl Popper, at the time a little known philosopher of science, published The Open Society and Its Enemies. He dedicated the book to the victims of both Hitler's and Stalin's camps and called it his “war effort.” The book had an enormous impact and spawned both imitators, such as Hannah Arendt's The Origins of Totalitarianism, and a great deal of debate. Whatever else it accomplished Popper's work politicized the history of ideas. Against the (...) devastation of the war Popper refused to grant either innocence or quietude to the history of philosophy. He argued that ways of thinking and talking had terrible consequences; that philosophers legitimated terrible damage in their seemingly abstract inquiry. (shrink)

Trends in the history of social science dedicated to the study of crime and punishment are presented as a case study supporting F.A. Hayek's theory of social change. Designing effective social institutions and public policies first requires an accurate vision of how society operates. An accurate model of society further requires scientific methods uniquely suited for the study of human beings as purposeful agents and the study of human institutions as complex social phenomena. If guided by faulty methods, theories are (...) inaccurate and policy outcomes veer from their intentions. Hayek termed such outcomes "abuses of reason". Aiming to replicate the objectivity of physical sciences via formal modeling and statistical measurement, economists throughout the 20th century imposed an excessively technical vision of human decision-making. Policy failures and social problems resulted. This paper argues that the historical trends of applied social science dedicated to crime and punishment can be understood similarly. Formal modeling and statistical measurement continuously displaced methods more attuned to human intentionality and social complexity. In result, amidst a long-run history of intellectual and political change, US law enforcement and criminal punishment policies became technocratic, and outcomes became disjointed from their stated intentions to promote social order and welfare. Antecedente. Se considera «eutanasia» al acto de acabar con la vida del paciente a petición expresa de él, para terminar el sufrimiento propio de alguna enfermedad sin perspectiva de cura. Objetivo. Evaluar los conocimientos y la percepción sobre la eutanasia en estudiantes y profesionales de medicina, demostrando los conocimientos y clasificando la percepción sobre la eutanasia. Metodología. Estudio clínico-epidemiológico, observacional, cualitativo-cuantitativo, de corte transversal y con muestreo no probabilístico por conveniencia, en los estudiantes y profesionales de medicina de un hospital (...) en Venezuela. Resultados. La muestra estudiada tuvo una distribución según grado de instrucción: 64.85% de estudiantes, 19.33% de residentes y 15.82% de especialistas. Presentaron un grado de conocimiento medio, distribuido en 84.4% de especialistas, 77.3% de residentes y 80.5% de estudiantes (p 0.051). De acuerdo con los grados de conocimiento y el apoyo a la eutanasia, el 76.18% tiene un grado medio de conocimientos (p 0.0002). Conclusión. Se demostró que existe un grado medio de conocimiento, en particular en la población estudiantil. La eutanasia no fue percibida de manera negativa, demostrando una alta significancia en la atención a las implicaciones éticas que rodean este concepto. (shrink)

The proportionality standard demands a meaningful link between the severity of crimes and the punishments received for them. This article investigates the compatibility between this philosophical demand and the practical means most commonly associated with criminal justice provision: governmental decision making. In so far as criminal justice systems require the coordination of real human and physical resources, certain forms of knowledge and incentives are required to calculate, produce, and distribute outputs proportionately. Whereas markets rely upon pricing mechanisms to generate and (...) coordinate information and incentives across diverse stages of complex production processes, governmental decision making often lacks a calculation mechanism comparable to market prices through which knowledge about societal needs and demands can be conveyed and may thus inevitably result in some forms of punitive disparity. John Searle’s argument that social-scientific laws are impossible depends on a special open-ended feature of social kinds. We demonstrate that under a noncontentious understanding of bridging principles the so-called "counts-as" relation, found in the expression "X counts as Y in (context) C," provides a bridging principle for social kinds. If we are correct, not only are social-scientific laws possible, but the "counts as" relation might provide a more perspicuous formulation for candidate bridge principles.
